Description: Nelfinavir mesylate (AG1343) is a potent HIV protease inhibitor. It is used in combination with other antiviral drugs in the treatment of HIV in both adults and children. AG1343 has potent antiviral activity (95% effective dose = 0.04 microgram/ml) against a number of HIV-1 strains in acute and chronic models of infection. AG-1343 is also an effective anti-cancer agent, with an EC50 of 3.1-5.2 micromolar and induced apoptosis and autophagic responses. AG-1343 also mediates AKT signaling demonstrating inhibition of endogenous and growth factor-induced Akt activation. AG1343 decreased the viability of a panel of drug-resistant breast cancer cell lines and inhibited the growth of NSCLC xenografts that was associated with induction of ER stress, autophagy, and apoptosis.
